چکیده

The effects of hydrogenated fats and butylated hydroxytoluene (BHT) in the diets of rats on the hepatic activation of benzo(a)pyrene, 2-acetylaminofluorene (AAF), and 2-aminofluorene by liver homogenates (S-9 fraction) were evaluated. The Salmonella/microsomal mutagenicity assay (Strain TA 98) was utilized to determine the mutagenic potential of the activated compounds. The S-9 fraction was obtained from animals fed a 15% fat diet consisting of hydrogenated fats (43% trans-fatty acids) or unsaturated fats (0% trans-fatty acids). BHT was administered orally (0.5%) 6 days prior to sacrifice in both groups. The incorporation of BHT in the diet of rats enhanced the mutagenic potential of AAF and 2-aminofluorene but not of benzo(a)pyrene. This effect was independent of the lipid composition of the diet. The most significant increment in the production of mutagenic metabolites was observed with AAF when BHT and hydrogenated fats were included in the diet of rats. Dietary hydrogenated fats appeared to potentiate the effects of BHT on AAF mutagenicity. Further studies to elucidate the mechanisms by which BHT and hydrogenated fats enhance AAF mutagenicity are warranted.

Effect of butylated hydroxytoluene on passive avoidance learning in male rats

Introduction: Butylated hydroxytoluene (BHT 2, 6-di-tert-butyl-p-cresol) is one of the extensively used antioxidants in the food industry. It is used in low-fat foods, fish products, packaging materials, paraffin, and mineral oils. BHT is also widely used in combination with other antioxidants such as BHA, propyl gallate, and citric acid for the stabilization of oils and high-fat foods. On t...

The role of nutritional lipids and antioxidants in UV-induced skin cancer.

Two dietary tenets of the free radical theory of cancer require refinement. The first was dietary reduction of vulnerable free-radical targets, e.g., polyunsaturated lipids. The second was the addition of one or more antioxidants to the diet. Further, it was reported in 1939 that high levels of dietary fat exacerbated UV-carcinogenesis. Both lines of enquiry (dietary lipid and antioxidant effec...

Effects of dietary fat on incidence of spontaneous and induced cancer in mice.

A study was done to evaluate the effect of quantity and degree of saturation of dietary fat on the incidence of spontaneous tumors in male and female Swiss mice and in female C3H mice and on the incidence of tumors induced by s.c. administration of 1,2-dimethylhydrazine (DMH) in Swiss mice. Diets contained 5 or 17% fat substituted isocalorically for sucrose in a semipurified diet. Gross autopsy...

Short-term effects of butylated hydroxytoluene on the Wistar rat liver, urinary bladder and thyroid gland.

Long-term feeding of butylated hydroxytoluene (BHT) to rats and mice has been linked to the enhancement of the incidence of liver tumors. It is shown in this paper that in the liver, urinary bladder and thyroid of the male Wistar rat, feeding the highest tolerated doses of BHT for 30 days does not lead to detectable increases in [3H]thymidine labeling. On the other hand, treatment of rats with ...

Effects of dietary constituents on ultraviolet light-mediated carcinogenesis.

The effects of several dietary supplements of antioxidants and enzyme inducers on ultraviolet light-mediated carcinogenesis were investigated. Glutathione (reduced) was without effect, but butylated hydroxytoluene, phenobarbital, and disulfiram all significantly suppressed the initiation and development of actinic lesions and tumors. On the basis of the present study and related previous ones, ...
